Analysis of the interaction between tryptophan-related compounds and ATP-binding cassette transporter G2 (ABCG2) using targeted metabolomics

ATP-binding cassette transporter G2 (ABCG2) is involved in the secretion of several compounds milk. The vitro and vivo interactions between tryptophan-related ABCG2 were investigated. tryptophan metabolome was determined by liquid chromatography-tandem mass spectrometry milk plasma from wild-type Abcg2?/? mice as well dairy cows carrying Y581S polymorphism (Y/S) noncarrier animals (Y/Y). milk-to-plasma ratios tryptophan, kynurenic acid, kynurenine, anthranilic xanthurenic acid higher than mice. ratio 2-fold Y/S Y/Y for kynurenine. In transport assays confirmed that some these substrates validated differences observed two variants bovine protein. These findings show metabolites belonging to kynurenine pathway into mediated ABCG2.
